Dr. Orville Kolterman is a pioneer in the diabetes space.

As Pendulum's Chief Medical Officer, Dr. Orville Kolterman is passionate about type 2 diabetes solutions that are both safe and effective.

Dr. Kolterman has had a prolific career in the medical field of type 2 diabetes. He was previously the Chief Medical Officer at Amylin Pharmaceuticals, where he brought four diabetes drugs from early-stage development through trials and to market. He also served as the President of the California Affiliate of the American Diabetes Association and is an Adjunct Professor of Medicine at the University of California, San Diego. Dr. Kolterman received his MD from Stanford University School of Medicine, performed his residency in Internal Medicine at the University of Chicago and returned to Stanford for his Endocrinology training.

What does your role entail as Pendulum's Chief Medical Officer?
I oversee all medical aspects of the company, which includes acting as the main source of medical advice, overseeing safety and monitoring activities for product clinical trials as well as answering medical questions about our existing product Pendulum Glucose Control from customers and physicians.

One of the most important facets of my role is to develop and run clinical trial programs for every product that we release. Since the very beginning, we've been committed to demonstrating clinical efficacy and safety for our products through rigorous scientific research and trials.  We have completed several preclinical and clinical assessments of efficacy and safety on Pendulum Glucose Control. PGC has also gone through DNA sequencing analysis, virulence factor analysis, antibiotic resistance testing, pathogen testing, and animal toxicity studies prior to any use in humans.

What makes Pendulum Glucose Control so different from other type 2 diabetes treatments? Why focus on a microbiome-directed solution?

The past few decades of academic research have documented that the amount of DNA that resides in the gut microbiome is more than a hundredfold greater than what is in the entire human genome. It's clear that there's a lot more complexity going on in the human gut and a lot more potential to unlock key interactions between the microbiome and the human genome. Just think of all the different combinations and permutations that you can put together. It's that interaction that's going to be the key to unlock the unknowns about a whole host of diseases.

For type 2 diabetes specifically, it's an opportunity to develop something that could help those affected without the common side effects of drugs. I was involved with bringing two new classes of diabetes drugs in the last 10 years, and they all come with significant side effects. Even now when I talk to friends who have diabetes practices, they share how their patients struggle with having to deal with the side effects of the new drug classes.  

With recent mounting academic and scientific research on the connection between the human microbiome and type 2 diabetes, there is more reassurance that beneficial bacterial strains provide a safer alternative as they've existed within the human gut for centuries. Specifically, the strains that we're working with and have incorporated into Pendulum Glucose Control have not been associated with significant side effects. We've also been extremely diligent in our selection of strains and producing our formulation through strain isolation and full characterization including complete sequencing of each bacterial genome. We've also tested each strain to ensure they don’t have any undesired antibiotic resistance or release toxins. It's been really exciting to have the opportunity to create a product that has proven to be effective and most importantly, safe.
